Hallo skysurfer

Denke du hast den Thread schon vollständig durchgelesen. Nun an der Situtation die da geschildert wird hat sich noch nicht viel geändert. Der jetzige Kurs wird im Yahoo-Forum folgendermassen erklärt:
- Shortys am Werk
- Seit Roche-Deal keine News mehr
- allgemeine Marktlage

Alle warten auf die Ergebnisse der Phase 1 Trials. Die Quartals-Zahlen bei Arqule sehe ich nicht als ausschlaggebend. Was zählt sind einzig und alleine die Phase 1 Trials. Im Yahoo-Forum haben einige herausgefunden, dass weitere Personen in die Trials einbezogen wurden. Dies wurde mit einer erfolgversprechenden Wirkung bei den ersten Testpersonen begründet. Die Trials werden an Personen durchgeführt welche im fortgeschrittenen Stadium eines Tumors sind. ARQ-501 hatte bei den Tierversuchen in Kombination mit Taxol über 90% der Tumore vernichtet. Unternehmen welche im Moment ein Medikament auf den Markt bringen welche das Leben von Krebspatienten um ein halbes Jahr verlängert, werden mit ca. 6 Milliarden Dollars bewertet (Arqule im Moment 150 Millionen Dollar). Tönt alles fantastisch, was aber nicht ausschliesst, dass ARQ-501 nicht funktioniert und die Investierten viel Geld verlieren.
Wenn die Phase 1 Trials erfolgreich sind, dann sind Kurse im 2-stelligen Bereich kein Problem, wenn nicht dann weiss ich auch nicht.

Nun es gibt keinen klaren Grund warum Arqule runter geht, ausser einige wissen mehr als wir (was zwar zwangsläufig der Fall ist, aber diese dürfen keine Aktien handeln, sonst ab ins Kittchen).

Das Volumen ist immer unter dem Durchschnitt der letzten 3 Monate, was nicht auf einen Sell-off schliessen lässt.

Nun die Frage wer verkauft? Hier gibt es einige Kandidaten die nach dem Roche-Deal im Hype gekauft haben und nun in der Panik verkaufen. Solche die auf ARQ-501 setzen (gehöre auch dazu) verkaufen sicher nicht nur weil der Kurs runtergeht (natürlich gibt es auch da eine Schmerzensgrenze) ohne dass Informationen gekommen sind.

Die Frage bleibt immer noch die gleiche, funktioniert ARQ-501, oder nicht.

Dies sind eigene Interpretationen gemischt mit Aussagen aus dem Yahoo-Forum.

08.07.2004 - Deutliche Verluste hat der Kurs von Arqule in den vergangenen Monaten verzeichnet. Auch am Mittwoch waren an der Nasdaq zwar Verluste in Höhe von knapp 1,7 Prozent zu verzeichnen, dabei hat sich die Aktie vom Tagestief aber deutlich erholt. Ausgebildet hat sich zu zwei Dritteln die Formation eines Morning Stars, der aber unbedingt am Donnerstag die Bestätigung durch eine weiße Kerze bedarf, um als potenzielle Trendwendeformation in Frage zu kommen. Auffällig sind die hohen Umsätze am Mittwoch.

Arqule: Potenzielle Trendwende nicht gelungen

09.07.2004 - Am Donnerstag hatten wir unsere Leser übe eine mögliche Trendwende bei Arqule informiert. Der Morning Star ist allerdings – siehe unsere gestrige Nachricht (hier kicken) – nicht zustande gekommen. Unbedingt notwendig war eine weiße Kerze am Donnerstag. Heraus gekommen sind Kursverluste, weshalb sich das Chartbild verschlechtert hat. Ein Rebreak über den gebrochenen Support bei 5 Dollar erscheint nun nicht greifbar, weitere Kursverluste wären damit aus rein charttechnischer Sicht programmiert.

Du kannst keine News erwarten bis die Phase 1 abgeschlossen ist. Dies ist auf ende Jahr terminiert. Also keine falschen Hoffnungen.

Das Vorgehen in der Phase 1 ist übrigens folgendermassen:
Die Dosierung des Medikamentes wird solange erhöht bis die toxische Schwelle erreicht wird. Einige auf Yahoo sehen es als positiv, dass es noch keine News gab (Phase 1 noch nicht beendet), was soviel heisst, dass die toxische Schwelle noch nicht erreicht ist, und die Dosis weiter gesteigert wird. Für ARQ-501 ist das gut wenn die toxische Schwelle möglichst hoch ist.

Wenn ARQ-501 fehlschlagen sollte, währe es nicht übertrieben, wenn das Unternehmen unter seinem CASH-Wert notiert, da keine Zukunftsaussichten bestehen.

Als aussenstehender ist im Moment kein schlechtes Anzeichen sichtbar. Hoffen wir somit auf eine erfolgreiche Phase 1.

Auf Dein Bitten habe ich mir den share mal angeschaut, zur Aktie selbst bzw. zum Fundament kann ich wenig sagen, außer, daß ich mitbekam, daß sie sehr gepusht wurde von zahlreichen Börsenblättchen im Rahmen des Nanohypes...zum Chart, die Aktie ist sehr überverkauft, könnte allerdings kurzfristig noch ein wenig tiefer rutschen, bitte achte drauf, daß der Chart in USD ist und nicht in Oiro ! ...ein Chart sagt mehr als "dausend" Worte ...die Channels sind bisher immer wieder gebrochen worden...

Tönt sehr positiv dieser Bericht.
Hier die wichtigsten Sachen:

- Know-How-Zuwachs mit Daniel D. Von Hoff
- DAS WICHTIGSTE: ARQ-501 scheint bis jetzt zu funktionieren, wie oben erwähnt ist die toxische Schwelle noch nicht erreicht. Es wird erste Trials im Einsatz in Chemotherapien geben. Es wird Trials geben im zusammenhang mit dem Medikament gemcitabine. Die erste Phase ist noch nicht abgeschlossen aber "continues to enroll successfully". Auf das letzte Quartal sind beginn der Phase 1b/2 geplant.
- Die Finanzmittel wurden im geplanten Rahmen verbraucht
- Arqule sitzt auf 76Mio Cash ($). Mit allen Besitztümmern kommt man auf ca 100Mio $. Dies macht auf die Aktie ca 3.5$, im Moment sind wir bei 4$ (noch).
- Es scheint als ob Arqule sich voll auf die Krebsforschung ausrichtet, da noch weitere Projekte erwähnt werden, welche im Zusammenhang mit Krebs-Medis gestartet wurden.

Ich muss sagen mit diesem Bericht fällt mir ein Stein vom Herzen, the show can go on.

Der Kurssturz in den letzten Wochen/Monaten (nach Roche-Deal) war tatsächlich fundamental unbegründet.

Eine Frage an die Spezis:

Wann wäre denn ein guter Zeitpunkt, um wieder in Arqule einzusteigen? Wenn ich mit meinem nachgewiesen limitierten Chartwissen auf den von f-h eingepflegten Chart (#90) schaue, so dürfte die obere Begrenzungslinie des Abwärtstrends heute etwa bei $ 4,25 liegen. In diesem Bereich ist Arqule nun auch notiert.

Fundamental sieht es auch passabel aus -unter Vorbehalt der bekannten und genannten Gefahren (wobei ich denke, dass Pfizer  und Roche nicht mit in ein seeuntaugliches Boot gestiegen wären).

Also liebe Leuts, the Show can go on.

In Genf war heute eine Krebskonferenz und Arqule hat auch präsentiert. Sie gaben erste Phase 1 Resultate bekannt. Als Info: Die Phase 1 dient alleine dazu die optimale Dosis eines Medikamentes herauszubekommen, das heisst ab welcher Dosis wirkt das Medikament toxisch und hat das Medikament in Menschen gefährliche Nebenwirkungen.
Es wurden Daten von 14 Personen erwähnt, wobei 3 Personen kein Tumorwachstum mehr hatten und bei einer Person der Tumor anfing zu schrumpfen. Die Dosis ist aber in den Menschen noch nicht bis an die Grenze gegangen und wir wissen auch nicht wieviele von diesen 14 Personen eine hohe Dosis erhielten.
Insgesamt nahmen 30 Personen an der Phase 1 teil. Es wurden also noch nicht alle Daten veröffentlicht. Am 5 Oktober ist eine weitere Präsentation in den USA. Somit besteht die Hoffnung, dass zu diesem Zeitpunkt weitere Resultate bekannt gegeben werden.
Zur Info: ARQ-501 alleine hat in Tieren nicht alle Tumore zum absterben gebracht (weiss leider die Prozentzahl nicht mehr, habe den Link zu den Daten verloren). Aber in Kombination mit Taxol hat es über 90% der Tumore in Tieren zum absterben gebracht. Die Phasen 1b/2 werden nun als Kombo mit Taxol und Gemcitabine ausgeführt.

"Our experience with ARQ 501 in a phase 1 setting has shown nothing to be inconsistent with our preclinical data - Unsere Erfahrungen mit ARQ-501 in der Phase 1 hat keine Inkonsistenz zu den Präklinischen (Tierversuche) Daten gezeigt" Dieser Satz dürfte doch ermutigend sein

On September 8, 2003, the Company acquired Cyclis Pharmaceuticals, Inc., an early stage cancer therapeutics company. The Cyclis acquisition provided the Company with the proprietary ACT platform, an oncology discovery pipeline, and ARQ 501, which is now in Phase I clinical trials. The ACT platform is an approach to cancer therapy that is intended to use small-molecule drugs to restore and activate cellular checkpoints that are defective in cancer cells and that, once activated, will cause the cancer cells to undergo programmed cell death. The ACT platform is based on the understanding that a therapeutic agent that reactivates the quality control, or checkpoint functions, of a cell has the potential to re-enable the cell to detect and respond to DNA damage.

Oncology Portfolio

The Company's oncology portfolio, based on the ACT platform, consists of four programs that it obtained through its acquisition of Cyclis. The first two seek to target and elevate E2F, a protein related to cellular checkpoint and cell death. The third and fourth involve different targets and mechanisms related to checkpoints and cell death pathways, namely Cancer Survival Proteins (CSPs) and p53.

ARQ 501 entered Phase I clinical trials in September 2003 and is the Company's first clinical-stage compound. ARQ 501 is aimed at causing rapid and sustained elevation in the checkpoint regulatory protein E2F1. In preclinical studies, ARQ 501 activated E2F-mediated checkpoints leading to selective cell death of cancer cells. The compound has demonstrated anti-cancer activity in mice when applied as both a single agent and in combination with chemotherapy. In contrast to its selectivity for tumor cells over normal cells, ARQ 501 is active against tumor cells with a broad range of genetic defects. ArQule's Phase I (monotherapy) dose-escalation study in patients with advanced solid tumors is underway at the Dana-Farber/Harvard Cancer Center in three Boston hospitals: Dana-Farber Cancer Institute, Massachusetts General Hospital and Beth Israel Deaconess Medical Center. The objectives of this study are to determine the safety profile (clinical tolerability) of ARQ 501 and a recommended dose to be used in Phase II clinical trials. Based on the results of its preclinical tests, which have shown ARQ 501 to be active against a large number of solid and hematological (bloodstream) tumors, the Company intends to investigate ARQ 501 in solid tumors both as a single agent and in combination therapy, and in hematologic tumors initially as a single agent.

Applying ArQule's platform in small molecule chemistry and intelligent drug design to its 550 series program, the Company is developing analogs and derivatives of ARQ 501. These new compounds are also aimed at modulating E2F. It has identified several such compounds, and is working to optimize these leads for further development by designing and selecting the version of a given lead compound that has the most advantageous set of drug-like characteristics possible.

In the Company's Cancer Survival Protein (CSP) program, it is developing compounds aimed at blocking cellular survival mechanisms that cancer cells possess, and thereby selectively triggering cell death in such cancer cells. CSPs are certain proteins, including cytosolic and nuclear proteins, that are inappropriately elevated to excessive levels in cancer cells. In an animal model of cancer, ArQule scientists have explored the feasibility of safely and effectively treating cancer by blocking the activity of cancer cell survival proteins with small molecule compounds.

In ArQule's p53 Modulation program, it is validating the newly discovered intracellular protein PUMA (p53-Upregulated Modulator of Apoptosis) as a target for treating cancer. p53 is a protein that controls several key cell cycle checkpoints. In normal cells, when p53 is present in sufficient amounts, the PUMA protein is produced. When PUMA is produced, it rapidly induces cell death in cancer cells. In human cancer, however, the gene that expresses the p53 protein is a commonly mutated gene. The result of this mutation is that the p53 protein is either not functional or not produced in sufficient amounts to perform its checkpoint-related functions, including the production of PUMA. In its PUMA program, the Company is researching a potential way to restore normal function to the p53/PUMA pathway by inducing the production of PUMA. ArQule is now developing cancer specific PUMA modulators. This program involves a different, but potentially equally important, checkpoint pathway to cell death, which may have application to certain types of cancer.

In the area of small molecule anti-cancer therapeutics, the Company competes with Ariad Pharmaceuticals, Genaera Corporation, Onyx Pharmaceuticals, OSI Pharmaceuticals, Oxigene, Inc., Telik Inc. and Tularik Inc.

Other Portfolio Programs

The Company has two internal drug discovery programs outside of its cancer focus, each of which was developed using its chemistry-based drug discovery technology platform. In order to focus on its cancer programs, it is exploring opportunities to out-license both of these programs.

In November 2003, the Company commenced GLP-toxicity studies with its lead compound, ARQ 101, a p38 MAP Kinase inhibitor for rheumatoid arthritis. Throughout 2003, it has progressed several compounds through advanced lead optimization demonstrating functional oral activity in a rat model of rheumatoid arthritis. In this established animal model, the data indicated that its compounds reduced joint swelling in a dose-dependent manner and were well tolerated at all doses studied.

ArQule's pain program involves the N-type Calcium channel for neuropathic pain. In the first quarter of 2003, the Company generated lead compounds with activity against the N-type Calcium channel and identified three families of chemicals. In the second half of 2003, in connection with its shift in resources to the oncology based programs, it held up this program just prior to beginning optimization work on the lead compounds.

Competitors acting both as chemistry service providers and as integrated drug discovery companies include Array BioPharma and Exelixis. Other competitors in the chemistry technology services market are Pharmacopeia, Inc., Albany Molecular Research, Inc., Evotec OAI and Discovery Partners International, Inc.
