Clinuvel: Lichtschutzmedikament Afamelanotide in letzter Phase vor Marktzulassung
Basis ist die Schweizer Phase III Studie

Das australische Unternehmen Clinuvel Pharmaceuticals Ltd (News/Aktienkurs) (GER:UR9)(ASX:CUV) hat an einer Phase III-Studie, die in der Schweiz durchgeführt worden ist, mit dem neuartigen Medikament Afamelanotide erste positive Zwischenergebnisse erzielt. Afamelanotide wird subkutan injiziert und entwickelt eine Wirkung über zwei Monate. Getestet wurden 14 Patienten mit der seltenen Stoffwechselkrankheit Erythropoetische Protoporphyrie (EPP). Die Testperiode dauerte 12 Monate. Weitere 87 Patienten befinden sich in Europa und in Australien in dieser Testphase. Diese Test werden voraussichtlich in der zweiten Hälfte 2009 abgeschlossen sein.

Was ist die Krankheit Erythropoetische Protoporphyrie (EPP)?

Menschen mit EPP leiden unter schwerwiegenden Symptomen, sobald sie Licht-/UV-Strahlung ausgesetzt sind. Diese Lichtunverträglichkeit (Phototoxität) entwickelt sich bereits im frühen Kindesalter. Meist sind langfristige, brennende Schmerzen und Blasenbildung auf Händen und im Gesicht die Folgen. In schweren Fällen kann es zu Leber-, Gallen- und Knochenschädigungen kommen.

Das neuartige Medikament Afamelanotide, das von Clinuvel entwickelt und getestet wird, bewirkt, dass die körpereigene Melaninproduktion in der Haut angeregt wird. Damit leistet Afamelanotide einen grossen Beitrag, sich vor gefährlicher UV-Strahlung zu schützen. Melanin ist bekannt für seine lichtabsorbierende sowie schützende Wirkung.

Die Studienergebnisse zusammengefasst

Die Ergebnisse basieren aufgrund der Tests an den ersten 14 Schweizer Patienten von insgesamt 101. Die 14 Probanten wurden in Höhenlagen in der Schweiz (stärkere UV-Belastung) getestet. Das Ergebnis zeigte eine statistische signifikante Verminderung der Schwere der phototoxischen Reaktion auf der Haut und eine erhöhte Melanindichte.

Bereits 2007 ergab die Phase III-Studie, dass die mit Afamelanotide behandelten Personen unter deutlich weniger schweren Symptomen litten als diese, die mit dem Placebo getestet wurden. Die Umweltbedingungen waren die gleichen.

Es gab bei der Studie keine durch das Medikament hervorgerufene, schwerwiegende oder unerwünschte Ereignisse.

Das Medikament ermöglicht es den Patienten ihre Lebensqualität erheblich zu verbessern. Auf ein Leben im Freien müssen die Patienten nicht mehr verzichten.

Die europäische und australische Phase III-EPP-Studie wird voraussichtlich in der zweiten Hälfte 2009 abgeschlossen sein.

Bedeutung der Ergebnisse für Clinuvel

Die heute bekanntgegebenen Resultate wertet Clinuvel positiv und verstärkt die Erwartung des Unternehmens, dass mit einer Marktzulassung des Medikaments Afamelanotide in den nächsten Monaten gerechnet werden kann. Die Ergebnisse rechtfertigen weitere Investitionen des Unternehmens, um die Studie mit einer noch grösseren Anzahl Patienten erfolgreich abzuschliessen.

Photodermatosen und UV-empfindliche Haut sind ein globales Problem. Clinuvel erhofft sich, das führende Unternehmen im Gebiet der Photoprotektion zu werden. Afamelanotide ist ein Wirkstoff mit vielen Anwendungsgebieten. Clinuvel konnte fünf durch UV und Licht verursachte Störungen identifizieren, bei denen Afamelanotide Anwendung findet und den Bedürfnissen der Patienten, die unter schweren und chronischen Symptomen leiden, dient.

Eine der schwersten Indikationen bei der Afamelanotide zum Einsatz kommt ist die Behandlung der organtransplantierten Patienten. Da diese lebenslang auf immununterdrückende Medikamente angewiesen sind, haben sie ein erhöhtes Hautkrebsrisiko.

Erwartungen für 2009

Clinuvel erwartet nach einem positiven Bescheid der FDA diese Tests auch in den Vereinigten Staaten durchzuführen. Der erfolgreiche Abschluss ist wichtig, um einer Vermarktung in den USA entgegenzusehen. Es wird erwartet, dass Dermatologen und eine Reihe von Ärzten im Bereich der Hämatologie, Onkologie und Organtransplantation auf die Wirkung von Afamelanotide im neuen Bereich der Photoprotektion aufmerksam werden und bereits in Phase III-Tests positive Effekte für ihre Patienten erzielen können.

Hi Pirat,

 

ich bin schon lange in Clinuvel investiert. News bekommt man nur spärlich. Die haben glaube ich eiin schlechtes Marketing. Die Kursentwicklung ist nicht so schlecht bisher. Allerdings geht das Papier meiner Meinung erst nach der Marktzulassung durch die Decke, jo na. Was mich nachdenklich stimmt ist das, dass alles sehr lange braucht. Phase II, Phase III, etc. und ich etwas die Gefahr sehe dass Investoren abspringen und das Unternehmen zu wenig Geld für die Finalisierung der Produkte hat. Ich bleib noch positiv, all zu lange wird es nicht mehr dauern...

European regulators to evaluate new drugs safety and efficacy for orphan disease erythropoietic protoporphyria (EPP)

MELBOURNE, Australia & BAAR, Switzerland--(BUSINESS WIRE)--
Clinuvel Pharmaceuticals Limited (ASX: CUV; XETRA-DAX: UR9; ADR: CLVLY) today announced that it has submitted a marketing authorisation application (MAA) for its first-in-class drug SCENESSE® (afamelanotide 16mg implant) to the European Medicines Agency (EMA). The MAA covers the use of SCENESSE® as a prophylactic treatment in adult patients with erythropoietic protoporphyria (EPP), a rare disease which causes absolute intolerance of patients skin to light.

SCENESSE®, which received an orphan drug designation for EPP in 2008, will be reviewed under the EMAs Centralised Procedure. An approval under this scheme will allow Clinuvel to market SCENESSE® in all 27 European Union member states as well as Norway, Iceland and Liechtenstein.

We are confident that we have provided sufficient data to demonstrate that SCENESSE® is a safe and clinically meaningful treatment for EPP, Clinuvels Chief Scientific Officer, Dr Hank Agersborg said. We firmly believe that our application will withstand the rigor of the regulatory review.

This filing is an important milestone in the evolution of Clinuvel, and is another landmark among the innovative therapies developed by the biotech sector, as afamelanotide is the first ever melanocortin filed for marketing approval, Clinuvels CEO, Dr Philippe Wolgen said. In the coming months we will continue our constructive dialogue with the regulatory authorities in order to bring this much-needed therapy to the EPP community across Europe.

Erythropoietic protoporphyria (EPP)

EPP is a rare genetic disease found mainly in fair-skinned people. It is characterised by severe phototoxicity (intolerance to light) of the skin resulting in intolerable pain, swelling and scarring, usually of exposed areas such as the face, hands and feet. Symptoms can vary from mild to extreme lasting pain requiring hospitalisation. Patients often lead an indoors and sheltered life, avoiding light and UV exposure to prevent symptoms. Presently there is no known effective treatment for EPP, which affects approximately 10,000 people globally, an estimated 4,000 in Europe.

During the Phase II and III studies in Europe, the US and Australia, SCENESSE® has been shown to enable EPP patients to expose themselves to (sun)light without incurring characteristic burns (phototoxicity). Pivotal trials (CUV029 and CUV030) showed SCENESSE® could reduce the severity of EPP symptoms and enable patients to lead more normal lives. A marked improvement in Quality of Life was also reported. Thus far no serious safety concerns have been identified from the use of afamelanotide in more than 650 patients involved in trials, including more than 250 EPP patients. SCENESSE® is being proposed as the first effective treatment to prevent phototoxicity in EPP.

Afamelanotide 16mg controlled-release formulation (SCENESSE®)

Afamelanotide, the active ingredient in SCENESSE®, is a linear peptide which activates eumelanin, the dark pigment, in skin. Eumelanin protects skin from light and UV radiation (photoprotection). SCENESSE® is administered underneath the skin as a dissolvable implant, approximately the size of a grain of rice, which activates eumelanin for a period of two months.

Summary
CUV announced that it has submitted a marketing authorisation
application (MAA) for its drug afamelanotide (16mg implant) to the
European Medicines Agency (EMA). The MAA covers the use of
afamelanotide as a prophylactic treatment in adult patients with
erythropoietic protoporphyria (EPP), a rare disease which causes
absolute intolerance of patients’ skin to light. Approval would allow CUV
to market afamelanotide in all 27 European Union member states as well
as Norway, Iceland and Lichtenstein. This news is in line with our
timelines. We expect a MAA decision in the EU in CY12. The EMA aims
to complete a MAA review within 210 days after submission.
High chance of success given positive results from pivotal
European Phase III study
In December, 2011, CUV announced that final analyses of its
confirmatory Phase III European study (CUV029) in erythropoietic
protoporphyria (EPP) have shown a clinically relevant, statistically
significant prophylactic treatment effect for patients who had been
administered its alpha-melanocyte stimulating hormone, afamelanotide
(16mg controlled-release formulation).
The primary objective of evaluating afamelanotide in EPP patients was
to determine whether the prophylactic effect has meaningful clinical
benefit. Afamelanotide treatment aims to allow patients to lead a life
which includes exposing themselves to ambient light and to engage in
outdoor activities. A similar, secondary objective was to assess the effect
of treatment on their Quality of Life (QoL).
The key results included:
 Patients receiving afamelanotide reported significantly less pain
associated with phototoxicity (median pain score 6.0, p=0.035);
 Patients on active drug experienced half as many phototoxic reactions
(p=0.044);
 Afamelanotide enabled patients to experience significantly more direct
sunlight exposure without pain (p=0.005); and
 Patients on active drug reported a greater improvement in their Quality
of Life (Day 270, p=0.011).
No safety concerns were identified during the study.

Thus it is logical that this day is chosen as the worldwide Rare Diseases Day: a day where extra attention is sought for many rare diseases and disorders from which adults and children suffer every day.

Often the causes of rare diseases aren’t known. There’s often no treatment (yet). That’s why the world needs Rare Diseases Day: to show that these patients too are entitled to care and treatment, like any other.

hallo mitaktionäre! weiss jemand was neues über die clinuvel aktie? übers internet ist es schwer an infos ranzukommen, hatte gedacht dass die aktie explodiert nach den 1.80 und richtig nach oben schiesst, aber jetzt sind wir wieder "unten" und es tut sich nix, wenn jemand aktuelle infos hat bitte posten, danke und ein schönes wochenende. gruss armer Hund
